The enormous physical-chemical heterogeneity within the metabolome presents the analyst with a problem – how measure the metabolome quantitatively with a biologically relevant sensitivity? This project aims at increasing metabolite coverage in metabolomics experiments through (1) combining several mass spectrometry ionization techniques and (2) selectively analyzing certain functional groups. Initial experiments (see figure) points towards multiple ionization mass spectrometry strategies to increase the coverage of the metabolome. We are currently exploring a combination of ionization approaches including ESI, APCI, APPI, MALDI and nanostructure initiated mass spectrometry (NIMS).
